In order to research the spherical spiral groove hybrid gas bearings, the Realizable k − ε turbulence model of gas film was established based on FLUENT. The simulation calculation method of 6-degrees of freedom passive grid was used, which can simulate the lubrication characteristics of the gas film transient flow field accurately. And the gas film pressure distribution and dynamic characteristic coefficients are numerically calculated. The dynamic and static pressure coupling effects of the gas flow field were analyzed, and the axis motion trajectory was simulated. The effect of rotation speed, gas supply pressure and tangential angle on the dynamic characteristic coefficients during bearing operation was analyzed. And the stability of the gas bearing was studied. The conclusion from the analysis shows that different rotation speed and gas supply pressure will change the pressure distribution of the gas bearing during the operation. The dynamic characteristics of the gas film can be changed by reasonably optimizing the operation parameters, which can change the whirl characteristics of the gas film and improve the stability. Through calculation and analysis, the tangential angle is selected between 55° and 60°, to ensure that the gas film has a high stiffness, while it also can obtain the larger damping. The simulation results and the experimental results are compared and analyzed to verify the correctness and effectiveness of the simulation method. At the same time, the research of this paper provided a theoretical basis for optimizing the bearing structure and operating parameters, improving the dynamic characteristics of gas bearings and improving the operation stability

According to the gas film force variation law, when the bearing axis is slightly displaced from the static equilibrium position, displacement and velocity disturbance relation expressions for the gas film force increment are constructed. Moreover, combined with the bearing rotor system motion equation, calculation model equations for the gas film stiffness and damping coefficients are established. The axial and radial vibration and velocity of the gas bearings during operation are collected. The instantaneous stiffness and damping coefficients of the gas film are calculated by the rolling iteration algorithm using MATLAB. The dynamic changes in the gas film stiffness and damping under different motion states are analyzed, and the mechanism of the gas film vortex and oscillation is studied. The results demonstrate the following: (1) When the gas bearing is running in the linear steady state in cycle 1, the dynamic pressure effect is enhanced and the stability is improved by increasing the eccentricity; when the gas supply pressure is increased, the static pressure effect is enhanced and the gas film vortex is reduced, but the oscillation is strengthened. (2) With the increase in rotational speed, the gas film vortex force gradually exceeds the gas film damping force, and the stability gradually worsens, causing a fluctuation in the gas film stiffness and damping, following which singularity occurs and a half-speed vortex is formed. Meanwhile, the gas film oscillation is intensified, and the rotor enters the nonlinear stable cycle 2 state operation. (3) As the fluctuation of the film force increases, the instantaneous stiffness and damping oscillation of the film intensifies, most of the stiffness and damping coefficients exhibit distortion, and the rotor operation will enter a chaotic or unstable state. Therefore, the gas bearing stiffness and damping variation characteristics can be used to study and predict the gas bearing operating state. Finally, measures for reducing the vortex and oscillation of the gas film and improving the stability of the gas bearing operation are proposed

Taking the hemisphere spiral groove hybrid gas bearings (HSGHGB) as the research object, the nonlinear dynamic lubrication analysis mathematical model of spherical hybrid gas bearings is established with the axis instantaneous position and instantaneous displacement speed as the parameters. The perturbation pressure control equation is solved by means of the finite difference method in generalized coordinate system. The calculation program is prepared based on VCþþ6.0, and the transient perturbation pressure distribution of three-dimensional (3D) gas film, nonlinear gas film force, and dynamic stiffness and damping coefficients are numerically calculated. The influences of different speeds, eccentricity ratios, and gas supply pressures on the dynamic characteristic coefficients of gas film are studied. The results show that the influence of bearing's supply pressure, speed, and eccentricity on the dynamic characteristics of gas film is significant. The dynamic equations of rotor-bearing system containing the gas film dynamic stiffness and the damping coefficients are established, and the stability of the gas film is predicted based on the Routh-Hurwitz stability criterion. The research provides the theoretical foundation for actively controlling the bearing running stiffness and damping and stemming the instability of gas film

Background/Aims: The activation of complement system and the formation of C5b-9 complex have been confirmed in the glomeruli of patients with mesangioproliferative glomerulonephritis (MsPGN). However, the role and mechanism of C5b-9-induced injury in glomerular mesangial cell (GMC) are poorly understood. Rat Thy-1N is an animal model for studying MsPGN. It has been revealed that the attack of C5b-9 to the GMC in rat Thy-1N is sublytic, and sublytic C5b-9 can cause GMC apoptosis, but the underlying mechanism is not fully elucidated. To explore the role and regulatory mechanism of C5b-9 in MsPGN lesion, we used rat Thy-1N model and first detected the change of microRNA (miRNA) profiles both in Thy-1N rat renal tissues (in vivo) and in the cultured GMCs with sublytic C5b-9 stimulation (in vitro). Then we determined the effect of miR-3546, which increased both in vivo and in vitro, on GMC apoptosis upon sublytic C5b-9 as well as the involved mechanism. Methods: Rat Thy-1N model was established and GMCs were treated with sublytic C5b-9. The rat renal cortex and the stimulated GMCs were obtained for miRNA microarray detection. Subsequently, the increased miRNAs were verified by real-time PCR. Meanwhile, to ascertain the ability of some miRNAs to upregulate cleaved caspase 3 and induce GMC apoptosis, the corresponding miRNA mimics were transfected into GMCs, followed by western blotting (WB) and flow cytometry mesurement. Thereafter, the miR-3546-targeted gene (SOX4) was predicted using bioinformatics approaches, and SOX4 expression in Thy-1N tissues and in the GMCs upon sublytic C5b-9 stimulation or miR-3546 mimic/inhibitor transfection were detected using real-time PCR and WB. To prove that miR-3546 can affect SOX4 gene transcription and SOX4 can regulate survivin expression, dual luciferase reporter assay, real-time PCR, WB and chromatin immunoprecipitation (ChIP) assays were performed. Furthermore, the role of miR-3546/SOX4/survivin axis in the GMC apoptosis induced by sublytic C5b-9 was examined using WB and flow cytometry. Results: Compared with normal renal tissues and untreated GMCs, there were 43 and 62 upregulated miRNAs (> 2-fold) in Thy-1N tissues and sublytic C5b-9-stimulated GMCs respectively. A total of 17 miRNAs were increased both in vivo and in vitro, 11 of which were validated by real-time PCR. Among them, miR-3546 could markedly promote GMC apoptosis and inhibit SOX4 or survivin expression in response to sublytic C5b-9, and either SOX4 or survivin overexpression markedly rescued the GMC apoptosis mediated by miR-3546 mimic. Additionally, SOX4 overexpression could reverse the survivin suppression by miR-3546 mimic, and SOX4 could bind to survivin promoter (-1,278 to -853 nt) and activate survivin gene transcription. Conclusion: MiR-3546/ SOX4/survivin axis has a promoting role in the GMC apoptosis triggered by sublytic C5b-9, and our findings may provide a new insight into the pathogenesis of rat Thy-1N and human MsPGN

Multimodal image fusion techniques can integrate the information from different medical images to get an informative image that is more suitable for joint diagnosis, preoperative planning, intraoperative guidance, and interventional treatment. Fusing images of CT and different MR modalities are studied in this paper. Firstly, the CT and MR images are both transformed to nonsubsampled shearlet transform (NSST) domain. So the low-frequency components and high-frequency components are obtained. Then the high-frequency components are merged using the absolute-maximum rule, while the low-frequency components are merged by a sparse representation- (SR-) based approach. And the dynamic group sparsity recovery (DGSR) algorithm is proposed to improve the performance of the SR-based approach. Finally, the fused image is obtained by performing the inverse NSST on the merged components. The proposed fusion method is tested on a number of clinical CT and MR images and compared with several popular image fusion methods. The experimental results demonstrate that the proposed fusion method can provide better fusion results in terms of subjective quality and objective evaluation

Some domestic scholars revealed the effectiveness of Wenshen Yiqi Keli (WSYQKL) on chronic obstructive pulmonary disease (COPD). However, the exact mechanism of WSYQKL on COPD is fuzzy and needs further research. We adopted UPLC-Q/TOF-MS to analyze the chemical components of WSYQKL. In in vitro experiments, human airway smooth muscle cells (hASMCs) were intervened with 2.5% cigarette smoke extract (CSE), medicine serum of WSYQKL, miR-155 mimic, and FoxO3a silencing. Cell viability, proliferation, migration, and the expressions of miR-155, PCNA, Ki67, p21, p27, and FoxO3a were examined by cell counting kit-8, EdU staining, Transwell assay, scarification assay, qRT-PCR, immunol cytochemistry, and western blot, respectively. The association between miR-155 and FoxO3a was assessed by database and luciferase reporter gene analysis. We identified 47 kinds of chemical compositions of WSYQKL in ESI+ mode and 42 kinds of components of WSYQKL in ESI− mode. The medicine serum of WSYQKL strongly alleviated the proliferation and migration of hASMCs induced by CSE in a concentration-dependent manner. The medicine serum of WSYQKL enhanced the levels of p21, p27, and FoxO3a and weakened PCNA and Ki67 levels in hASMCs induced by CSE with the increase of concentration. MiR-155 mimic or FoxO3a silencing notably advanced CSE-treated HASMC viability, proliferation, migration, and the levels of PCNA and Ki67 and downregulated the levels of p21, p27, and FoxO3a in CSE-triggered hASMCs, which was reversed by WSYQKL-containing serum. Our results described that WSYQKL alleviated the proliferation and migration of hASMCs induced by CSE by modulating the miR-155/FoxO3a axis
